The Applied Biosystems HIV-1 Genotyping Kit features:

  • A standardized, cost-effective assay in a scalable workflow
  • A total workflow of about 14 hours for processing 14 samples from extracted viral RNA to results, with total hands-on time of less than 3 hours
  • The ability to detect mutations in the protease and reverse transcriptase regions of the HIV-1 pol gene in subtypes A, B, C, D, AE, CRF01_AE, CRF02_AG, and AG
  • Easy-to-read sequencing results with free software for assembly and interpretation

Step 1 > Amplification module

proflex  After sample processing and RNA extraction, samples undergo an initial reverse transcription + PCR, followed by a second nested PCR step for mutation targets of interest. The HIV-1 Genotyping Kit Amplification Module includes the RT-PCR master mix, the nested PCR master mix, and necessary positive and negative controls that allow for a simple, streamlined workflow as you move into your cleanup and sequencing steps.

Step 2 > Sequencing module

3500xl  Once you confirm and clean up your targeted PCR product, the HIV-1 Genotyping Kit Sequencing Module is used for cycle sequencing. Included in this kit are the forward/reverse pre-formulated sequencing mixes for the targets of interest and the pGEM sequencing control.

Step 3 > Data analysis and results

Free access to Exatype™ software is provided with the purchase of HIV-1 Genotyping Kits.  This easy-to-use application allows for:

  • Data upload directly to a secure web interface
  • Analyzing sequencing data files, creating consensus for target regions and reporting drug resistance into a user-friendly report
  • Integration with laboratory information management systems (LIMS)
  • Multiple users under a single organization license

The Applied Biosystems HIV-1 Genotyping Kit harnesses gold-standard Sanger sequencing technology to amplify and reliably sequence the diverse and rapidly evolving HIV-1 virus. To address the HIV surveillance needs of the global community, this kit offers broad genotyping coverage of HIV-1 Group M subtypes from plasma and dried blood spot (DBS) samples to detect resistance to protease inhibitors, nucleoside reverse-transcriptase inhibitors, and non-nucleoside reverse-transcriptase inhibitors.

This improved Applied Biosystems assay workflow was developed by Thermo Fisher Scientific in collaboration with the US Centers for Disease Control and Prevention.
